To be off is a casual way to say that they are leaving.

Like if you're having breakfast with your family and then say, okay, I'm off to work.

So Jo hates the fact that she's staying behind while everyone else is moving on with their lives.

She feels restless or anxious and she's not satisfied with her current situation.

She jokingly suggests running off to a pirate ship, meaning she wishes she could leave it all behind and go on an adventure instead of facing reality.

To run off means to leave quickly or suddenly.
